The magnitude and temporal pattern of insulin secretion is determined by interplay between several intracellular messengers and signalling proteins in pancreatic b-cells. Live-cell imaging have shown that glucose and neuro-hormonal stimuli induce dynamic changes of the sub-plasmalemmal concentrations of e.g. ATP, Ca2+ and cyclic AMP, which contribute to pulsatile insulin secretion. Moreover, secretory products elicit auto- and paracrine signalling that influence the secretion pattern. This presentation will review some recent insights into the spatio-temporal regulation of these signalling systems and how signalling defects may contribute to impaired insulin secretion in type 2 diabetes.

Major part of a pancreatic islet is composed of beta cells that secrete insulin, a key hormone regulating influx of nutrients into all cells in a vertebrate organism to support nutrition, housekeeping or energy storage. Beta cells constantly communicate with each other using both direct, short-range interactions through gap junctions, and paracrine long-range signaling. However, to what extent each of these cell-cell interactions shape collective sensing and cell behavior in islets that leads to insulin release is largely unknown. The functional multicellular imaging (fMCI) records a full temporal activity trace for every cell in an optical plane of an islet from which meaningful quantitative statements about the dynamics of unitary Ca2+ responses and information flow in the beta cell collective are possible. I will present how computational methods from network analyses, spin glass model to random matrix theory form valuable tools to extract the mechanisms underlying collective sensing and behavior of beta cells in pancreatic islets.

A one-day open international symposium on diabetes related research is organized annually at KI campus. The aim is to increase interactions between internationally renowned researchers within diabetes and metabolism and the research community at KI and in Sweden.
Attendance is free of charge and open for everyone. However, registration is mandatory and is made via email to Barbro.Svensson@ki.se. Last day for registration is November 1st.

Professor Raul Cavalcante Maranhão, University of São Paulo, Brazil
Professor Maranhão graduated in Medicine at the University of Brasilia (1973) and completed his Residency in Clinical Medicine and Endocrinology, at the Hospital das Clínicas, Faculty of Medicine, University of São Paulo (1975) and Doctorate in Sciences (General Physiology) at the Institute of Biomedical Sciences, University of São Paulo ). He has a postdoctoral degree from the Biophysics Institute, Boston University Medical School (1982-1984) and was a Visiting Professor at the Dept. of Molecular Genetics of that University (1990). He has a PhD from FCF-USP (1987), in the area of Clinical Biochemistry. He is currently a Full Professor at the Faculty of Pharmaceutical Sciences at USP and directs the Laboratory of Metabolism of Lipids of the Heart Institute (InCor) of the Hospital das Clínicas of the Faculty of Medicine of USP. His main lines of research are related to the metabolism of lipoproteins and use of nanoparticles for drug delivery for the treatment of degenerative diseases such as cancer and cardiovascular diseases.
